Oh, yes, I am sure it can! It has for me, many times. For example, one time I was feeling extremely stressed - I was making a transition to another position within the company for which I worked, and was basically (at least was trying!) to work both positions for the time being. These positions were conflicting in needs - one was very active, almost hectic pace and of the moment; the other (the new one) demanded quiet time and thought. I knew I was not giving the new position the attention it deserved and that I wanted to give it. Just couldn't seem to until I could move on from the other position! Frustration, stress, etc. set in.
One day I really called out to the Divine for help - wholeheartedly. The message I got back was a quote from Mary Baker Eddy's book Science and Health - "We are all capable of more than we do." I just laughed! Here I felt like I had a plateful to do, and God is telling me I am capable of doing more! That idea and the laughter it generated - a powerful spiritual quality in my book - broke the stress for me. After that, the stress and frustrations were gone and I was able to find ways to better handle the needs of each of the positions. It was still a month or so before the transition was resolved (and that by my being offered another position I never dreamed of but that really fit my abilities), but I enjoyed the time so much more.
So, small example, but yes! spirituality can relieve this burden you feel and help you find happiness again.
